Kinase proteins are intimately involved in cell signalling, and in many cases mutations to these proteins are involved in the development of cancers. A number of small-molecule drugs have been developed to target these proteins, most notably gleevec, which binds to the BCR-ABL kinase and is used as a treatment for chronic myelogenous leukemia, and iressa, which binds to the epidermal growth factor receptor’s (EGFR) tyrosine kinase domain and is used to treat non-small cell lung cancer. However, as is the case for bacteria and viruses, the tumour proteins can mutate and thereby develop resistance to these drugs. The mechanisms by which these mutations confer drug resistance are unclear, and are likely to depend on the particular mutation involved. Clearly, however, if we are able to understand these mechanisms, we are then in a position to design novel inhibitors which are unaffected by these mutations.

In this studentship, you will address the issue of how mutations of EGFR confer drug resistance to kinase inhibitors by using a range of simulation techniques, including docking and scoring, fast free energy calculations, and more accurate rigorous free energy methods. Methods to calculate the relative free energies of different protein conformations will also be used. This studentship is funded by the EPSRC and AstraZeneca (AZ), and will be supported by direct access to relevant experimental data available through AZ. The project will be supervised by Prof Jonathan Essex at the University of Southampton, together with a senior scientist from AZ. You will be joining an established research team working in the area of applying and developing computer simulation methods to biological problems.

Applicants should have, or expect to receive shortly, a good degree in chemistry, physics, biochemistry, or a relevant discipline, and a keen interest in molecular modelling. Please contact Dr Richard A Ward (richard.a.ward@astrazeneca.com) for enquiries related to this project and for further information regarding the nature of this industrially- sponsored studentship.

Industrial Supervisor: Dr Richard A Ward, AstraZeneca, Alderley Park, Macclesfied.

Universität Stuttgart with its 20,000 students and 7,000 employees is one of the leading European Research Universities with a main focus on engineering and natural sciences. Universität Stuttgart is committed to excellent basic and applied research and interdisciplinary academic education. The excellence initiative of the German state and federal governments has approved the funding of the Cluster of Excellence EXC 310 Simulation Technology (SimTech) at Universität Stuttgart (www.simtech.uni-stuttgart.de). In this context, the newly established Stuttgart Research Centre for Simulation Technology (SRC SimTech) is asking for applications for a

PhD student for the project Molecular Modelling of Enzymes at a Water-Substrate Interface

to pursue research in future-orientated research fields.

Lipases und PHA depolymerases are interesting enzymes for many biotechnological applications. While they are water soluble, they convert water-insoluble substrates by binding to the water-substrate interface. Our goal is to understand the molecular basis of the different substrate specificities of lipases and PHA depolymerases which convert triglycerides and polyesters, respectively. In this project, the molecular mechanism of binding will be studied by molecular dynamics simulations of large, realistic protein systems to understand the effect of the interface to structure and dynamics of the enzyme, and to design enzyme variants with improved biochemical properties in collaboration with our experimental project partners.

Candidates with a degree preferably in biology or chemistry and interested in an interdisciplinary and application oriented project at the interface between life science and engineering are invited to send their application to Prof. Dr. Jürgen Pleiss (Juergen.Pleiss@itb.uni-stuttgart.de), Institute of Technical Biochemistry, Allmandring 31, 70569 Stuttgart, Germany.

Postdoc and PhDs are expected to participate actively in the SimTech Teaching Programmes and Industry Transfer Concepts.

Applications with the usual documents (CV, degree certificates, references, publication list, letter of motivation) should be sent directly to the Professors/Junior Professors to arrive not later than April 30, 2009.

Universität Stuttgart wishes to increase the proportion of female academic staff and, for this reason, especially welcomes applications from women. Severely challenged persons will be given preference in case of equal qualifications.

SINGA is an award given to international students with excellent academic undergraduate and/or master’s results, and strong interest in doing research leading to a doctorate (PhD) in Science and Engineering at a Singapore University. The students can choose to do their research work mainly at the Agency for Science, Technology and Research (A*STAR), National University of Singapore (NUS), or Nanyang Technological University (NTU). The PhD degree will be conferred by either NUS or NTU.

“International students” refer to students who are not citizens or permanent residents of Singapore, and who have not previously studied in Singapore. We hope to award up to 240 SINGA scholarships per year with no bond. You are not required to take up citizenship and you are not required to work in Singapore after graduation.

Singapore International Graduate Award (SINGA)
A unique opportunity to pursue your PhD in Science and Engineering

The Singapore International Graduate Award (SINGA) is a collaboration between the Agency for Science, Technology & Research (A*STAR), the National University of Singapore (NUS) and the Nanyang Technological University (NTU). PhD training will be carried out in English at your chosen lab at A*STAR Research Institutes, NUS or NTU. Students will be supervised by distinguished and world-renowned researchers in these labs. Upon successful completion, students will be conferred a PhD degree by either NUS or NTU.

“We want to encourage more such exchanges, to build friendships and mutual understanding. This is why we are introducing a new Singapore International Graduate Award (or SINGA), for international students to do their PhD training in a multi-disciplinary environment in Singapore. 240 SINGA Awards will be given every year to top students from all over the world. I hope that this will enhance the close educational ties between our countries and will bring our young talents even closer together.”
